چکیده
Traditional treatments of parasitic and bacterial diseases are based on chemotherapeutic compounds such as chloramine-T and CuSO4. Although many compounds are used in fish treatments, their undesirable effects are not known. In this study, glucose 6-phosphate dehydrogenase (G6PD) was purified from rainbow trout (Oncorhynchus mykiss) erythrocytes by hemolysate preparation, ammonium sulfate precipitation and 2', 5'-ADP Sepharose 4B affinity gel chromatography in a single day. The enzyme, with a specific activity of 14.51 EU/mg protein, was purified 906.8-fold with a yield of 70.38%. To check the purity of the enzyme, SDS polyacrylamide gel electrophoresis was performed, which showed a single band. The effects of chloramine-T and copper sulfate (CuSO4) on the G6PD were investigated in vitro. Chloramine-T and CuSO4 had inhibitory effects on the enzyme. I50 values of the chemotherapeutic compounds were determined by plotting percent activity and Ki values, and types of inhibition were determined for each compound by means of Lineweaver-Burk graphs. In vivo studies showed that G6PD in rainbow trout erythrocytes was significantly inhibited by CuSO4 in one hour but not inhibited by chloramine-T.

High density lipoprotein loses its effect to stimulate efflux of cholesterol from foam cells after oxidative modification.
In this study, we performed oxidative modification of high density lipoprotein (HDL) in vitro. The amount of lipid peroxide increased when either HDL2 or HDL3 was incubated with phosphate-buffered saline containing 5 microM CuSO4 for 24 h at 37 degrees C, indicating that both fractions of HDL were oxidatively modified. This modification resulted in denaturation of apolipoprotein AI on SDS/PAGE ...
متن کاملEvaluation of the efficacy of d-penicillamine and trientine as copper chelators using an in vitro technique involving ovine red blood cells.
An in vitro technique for haemolysing ovine red blood cells with copper sulphate was standardized to induce c 50% haemolysis with 0.5 mM CuSO4 after incubation for 14 h at 38 degrees C. This technique was then applied to test the efficacy of trientine and d-penicillamine in preventing haemolysis. Trientine concentrations of 0.5; 1.0 and 1.5 mM were found to be the most effective (P < 0.05) in r...

Keratin was extracted from purified cornified cells of newborn rats in Tris-HCl-buffered 8 M urea containing beta-mercaptoethanol. Microfilaments were assembled in vitro by reducing the ionic strength of buffer and the urea concentration. One millimolar concentration of KCl and NaCl did not affect filament formation, but the same concentration of divalent cations greatly altered this process. C...

Dear Editor, Honey, a dietary source of natural antioxidants1, could have a protective role against diseases, such as cancer and cardiovascular diseases2,3. The present study is an attempt, the first to our knowledge, to examine the effects of Greek pine tree honey on the in vitro oxidation of human serum lipoproteins and isolated plasma low density lipoproteins (LDL). Serum and isolated LDL4 w...
متن کاملContribution of copper binding to the inhibition of lipid oxidation by plasmalogen phospholipids.
The role of plasmalogen phospholipids for copper-induced lipid oxidation was evaluated. Using 1H-NMR we observed that the copper (CuSO4)-promoted oxidative degradation of polyunsaturated fatty acids in micellar solution was dose-dependently attenuated by the plasmalogen lysoplasmenylethanolamine from bovine brain (lysoBP-PtdEtn). This was due to a direct interaction of copper ions with the plas...
